Nature of Genetic Material- Discovery of Nuclein

Chemistry of genes have their origin in Germany, early on in 1869 by Friedrich Miescher.  Miescher began his quest on genes by isolating nuclei from pus cells (white blood cells) in waste surgical bandages.  He had found that the nuclei that he had isolated contained a novel phosphorus-bearing substance that he decided to name nuclein.  Now we know that nuclein is mostly chromatin, which is a complex of DNA and chromosomal proteins.

Towards the end of the 19th century, both DNA and RNA were separated from the protein that clings them to a cell.  This then further allowed more detailed chemical analysis of these nucleic acids.
